چکیده

Background: Exosomes in synovial fluid (SF) has a close relationship with the pathogenesis of rheumatiod arthritis. As complex biological fluid, SF presents challenges for exosomes isolation using standard methods, such as Exoquick TM kit and ultracentrifugation. Objectives: The study aims to compared quality separated by (TM), kit+Exoquick TC (TM-TC), ultracentrifugation (UC) TM-TC+UC(TM-TC-UC) from SF. Methods: was TM, TM-TC, UC TM-TC-UC respectively. size concentrations were detected high sensitivity flow cytometry nanoparticle analysis. Total protein RNA extracted exosomes. SDS-PAGE used detect distribution Western blot examine level albumin marker (TSG101 CD81). Results: There no statistic difference diameters four methods. (5.65±0.93), (3.02±1.19), (1.67±0.25) (4.61±0.73) *10 9 Particles/mL. methods consistent showed that different. Low levels TM-TC TM-TC-UC, while UC. higher than other groups. Conclusion: can be obtain exosome-enriched components. References: [1]Helwa I, et al, A Comparative Study Serum Exosome Isolation Using Differential Ultracentrifugation Three Commercial Reagents. Optimization of The Cell Aggregates Method for Isolation and Purification of Human Granulosa Cells from Follicular Fluid

Objective Aspirated ovarian follicular fluids (FF) contain luteal granulosa cells (LGC) and other contaminating cell types. Several strategies such as the antibody binding methods, the flask method, the cell strainer, and the positive selection of granulosa aggregates after density gradient centrifugation, were tested as LGCs purification methods. Each of these strategies has its own advantages...

A method for counting monosodium urate crystals in synovial fluid.

This study was aimed to standardize the technique for counting monosodium urate (MSU) crystals in the synovial fluid (SF) of patients with gout. A total of 52 SF specimens were examined under a polarized light microscope. The amount of SF ranged between 0.1 and 45 mL (median 3 mL). MSU crystals were counted in four areas with the same size at 400x magnification. Cytological examination of the s...

Isolation of varicella zoster virus from the synovial fluid of a patient with herpes zoster arthritis.

Only a few cases of herpes zoster-associated arthritis have been reported to date (1-3). This form of arthritis is presumed to be viral in origin. Isolation of varicella zoster virus (VZV) by culture from synovial fluid has not been previously documented. VZV infection of the joint space associated with acute arthritis has been evidenced only once, by the finding of VZV antigen in the joint flu...

[Synoviocyte cultures from synovial fluid].

UNLABELLED The study of the pathogenetic mechanisms of rheumatic diseases is in general carried out through "in vitro" systems based on cellular cultures models. The difficulties to achieve fresh human tissue prompted us to develop a simpler method to obtain fibroblast-like synovial cells from synovial fluid (SF). METHODS SF was collected from the knees of 5 patients with rheumatoid arthritis...

A novel method for endothelial cell isolation.

The present study aimed to develop a quick and efficient method for purification of newborn endothelial cells from tumor tissues. Fresh tissues were separated from C57BL/6 mice bearing tumors derived from mouse lung cancer Lewis cells, fully minced and divided into two parts. One part was subjected to collagenase type I digestion with a vortex to form a single-cell suspension, while another par...
